Azo switch is widely used as an important component of optical response system.
On October 7, 2024, Yang Guangfu and Yin Jun of Central China Normal University jointly communicated inNature Communications Published online entitled“A visible light-activated azo-fluorescent switch for imaging-guided and light-controlled release of antimycotics”The research paper,TheAn azo fluorescence switch was developed, which is visible light responsive, and its light response process can be monitored by fluorescence imaging.
Visible light irradiation promotes isomerization, accompanied by a change in fluorescence, enabling this process to be monitored by fluorescence imaging. In addition, we recorded the change in the nanocavity size of azo-containing liposome-encapsulated nanoparticles during isomerization, and showed that this change can construct a light-responsive nanoplatform for optically controlling the release of antifungal drugs. At the same time, natural light activation of switch nanoparticles with antifungal agents resulted in the death of Rhizoctonia blight. The results show that the retention period of these nanoparticles can be doubled compared to small molecule antifungal drugs. The strategy for designing an imaging-guided light-controlled nano-antifungal release system can be applied to a variety of drug controlled delivery protocols.
Azo compounds are commonly used as molecular switches in materials science because they are capable of reversible isomerization between the E and Z forms under the action of light.Due to its non-invasive and biocompatibility, the potential application of visible light-activated azo optical switches in biological systems has received widespread attention. In particular, azo light switches have been developed for engineering biomolecules, such as proteins and nucleic acids, so their function can be controlled by visible light-induced conformational changes. In recent years, azo-switch-mediated optical control has been used to explore and manipulate various cellular processes. The changes in molecular structure and the regulation of light on cell behavior are mainly manifested at the level of cell microenvironment. It should be possible to design new types of azo switches that coordinate the optical control of biological processes with fluorescence imaging techniques for optical tracking and monitoring of dynamic processes. Therefore, the E-Z photoisomerization of the new azo fluorescent molecular switch designed using this strategy can not only modulate biological processes with light, but also induce fluorescence changes to signal transduction and evaluate photomodulatory events.
A typical strategy for constructing azo fluorescent molecular switches involves the direct introduction of a fluorophore at the end of the azo unit or azobenzene moiety. However, fluorophore-modified azo compounds are generally susceptible to fluorescence quenching, which is caused by fluorescence resonance energy transfer (FRET) between the fluorophore and the azo unit. In addition, the introduction of fluorophores can have a deleterious effect on photoisomerization efficiency and often cause cytotoxicity and bioincompatibility. In addition, azo compounds having a fluorophore directly attached to an azo group are difficult to prepare by simple synthetic methods. The direct incorporation of azoarene groups as platforms for azo fluorescent molecular switches may be able to avoid these problems. However, it is well known that azoarenes undergo non-radiative processes caused by rapid internal conversion, resulting in energy dissipation in the form of heat rather than luminescence.

In considering alternative strategies that might circumvent these problems, studies were concerned with the design of visible-light-activated azo-fluorescent molecular switches containing azoarene N = N double bonds as conjugated bridges separating electron donors and electron acceptors. The study suggests that this approach can create a donor-acceptor (D-A) system. The study evaluated the optical switching performance of visible light-activated azo fluorescent molecular switches in vitro and in vivo. It is demonstrated that the E,Z photoisomerization of this photoactivated switch in the cellular microenvironment can be monitored by fluorescence imaging. The structural change promoted by the photoisomerization of this azo fluorescent switch is used as the basis of a nanoplatform for the photocontrolled release of drugs.
