The Principle and Application of Raman Spectroscopy

Author:

Principle of 1. Raman Spectroscopy

Raman spectra is a kind of scattering spectrum. Raman spectroscopy is based on the Indian scientist C.V. Raman (Raman) found the Raman scattering effect, and the incident light frequency is different from the scattering spectrum analysis to obtain molecular vibration, rotation information, and applied to the molecular structure of an analysis method.

Raman is a light scattering technique in which light is scattered elastically and inelastically when it hits a substance. The scattered light of the elastic scattering has a component having the same wavelength as that of the excitation light, and the scattered light of the inelastic scattering has a component having a wavelength longer and shorter than that of the excitation light, which is collectively referred to as the Raman effect.

On the coordination of the vibration, the change of the molecular polarization potential or the change of the electron cloud is the result of the molecular Raman effect. The amount of change in polarizability will determine the Raman scattering intensity. The change in the mode frequency is determined by the rotational and vibrational state of the sample.

1.Rayleigh scattering: elastic collision; no energy exchange, only change direction;

2.Raman scattering: inelastic collision; direction change and energy exchange;

Applications of 2. Raman Spectroscopy

Raman spectrometer is a kind of spectral analysis instrument, which is widely used in many fields. Here are some of the main application areas:

1. Chemical and Physical Studies

Raman spectroscopy can provide structural information of substances, which is suitable for the study of chemistry, physics and other disciplines.

In catalytic chemistry, Raman spectroscopy is used to study the structure of catalysts and their surface species, as well as real-time studies of catalyst preparation processes.

In the field of polymer materials, Raman spectroscopy is used to analyze the structure of polymer materials, such as molecular structure, stereoregularity, crystallization and destination.

2. Artistic and Cultural Heritage Identification

Raman spectroscopy was used to identify red dyes such as safflower and yellow dyes such as sophora dyes in textile artifacts.

3. Applied to the identification of the authenticity of works of art, such as the identification of gems.

4. Criminal investigation and geology

In criminal investigation, Raman spectroscopy is used to analyze physical evidence at a crime scene.

In geology, Raman spectroscopy is used to analyze the composition and structure of minerals.

5, Life Sciences

Raman spectroscopy can be used for the structural analysis of biological macromolecules such as proteins and nucleic acids.

In medical diagnostics, Raman spectroscopy can be used to detect changes in the concentration of specific molecules within cells.

Other areas

Raman spectroscopy is also used in the field of QC, providing fast and simple non-destructive qualitative and quantitative analysis.

In the fields of carbon materials and semiconductors, Raman spectroscopy is used for in-situ analysis to provide structural information of materials.
